Description

The present invention relates to new psoralen modified Nucleotide derivatives, their preparation and their use.

Psorale (Furocumarine) stellen eine natürlich vorkommende Klasse heterocyclischer Verbindungen dar (P. S. Song und K. J. Tapley, Photochem. Photobiol. Psorals (furocoumarins) represent a naturally occurring one Class of heterocyclic compounds (P. S. Song and K. J. Tapley, Photochem. Photobiol. 29 [1979], 1177-79), the due to its planar, aromatic structure Intercalation to double-stranded areas of DNA and RNA can bind.

J. Med. 316 [1987], 297-303).The psoralen are bifunctional, photoreactive Compounds that when exposed to near UV Light photocycloadditions can enter, preferably Pyrimidine bases. The addition takes place in a 2 + 2 cycloaddition, either with the furan (5′-4 ′) double bond or the Pyron (3-4) double bond of the psoralen framework, or, at adjacent arrangement of the pyrimidine bases, with both double bonds; therefore they are both monoadducts as well as "interstrand crosslinks" possible (D. Kanne et al., J. Am. Chem. Soc. 104 [1982], 6754-64). The photoaddition takes place with high preference on thymidine or uridine bases (E. Garrat-Wheeler et al., Nucleic acids res. 12 [1984], 3405-3423). Because of their high specificity to nucleic acids, easy uptake into biological systems as well of their photochemical properties that the "freezing" enable double-stranded nucleic acid regions, are suitable Psoralene as an ideal tool for studying secondary DNA and RNA structures (see C.-K. J. Shen et al., Proc. Natl. Acad. Sci. USA 73 [1976], 2649-53). Besides this biochemical area of application will be psoralene derivatives for a long time with great benefits in clinical Area for the treatment of psoriasis and similar skin diseases used (see J. H. Parrish et al., N. Engl.  J. Med. 291 [1974], 1207-1211), as well as more recently successful in photochemotherapy of cutaneous T cells Lymphoma, a skin cancer that has been almost incurable to date (R. Edelson, N. Engl. J. Med. 316 [1987], 297-303).

Natl. Acad. Sci. 83 [1981], 1127-1231).In recent years, especially in the With regard to the clinical-medical application in the AIDS therapy, an increasing interest in the use of so-called "Antisense" oligonucleotides. This means Deoxy or ribonucleic acids, the sequence of which ranges of messenger RNA's is complementary and that is through binding translation to the sequence sections, both in vivo block efficiently as well as in vitro (cf. Y. Shi et al., Nucleic acids res. 15 [1987], 6843-6854). To To increase the nuclease stability, the phosphate Internucleotide binding in different ways modified, the methylphosphonates being particularly noteworthy (P. S. Miller et al., Biochem. 25 [1986], 5092-5097) and the thiophosphates (T.A. Kunkel et al., Proc. Natl. Acad. Sci. 83, 1981, 1127-1231).

U. Asseline et al., Nucleosides and Nucleotides 5 [1986], 45-63). 27 [1988], 3197- 3203).To increase hybrid stability, especially for short ones Oligonucleotide sequences, and thus to increase the inhibition efficiency, intercalating reagents such as e.g. B. coupled the acridines to the termini of the oligonucleotides (See U. Asseline et al., Nucleosides and Nucleotides 5 [1986], 45-63). It has been found to be particularly advantageous proved the coupling of the psoralens to oligonucleotides. Several working groups have shown that it it is possible to covalently modify such modified oligonucleotides and thus irreversibly to the desired target sequence bind in this way to the most efficient blocking possible to achieve translation. The coupling of the psoralen remains can be used as a so-called "site specific engineered" Monoadduct (see, for example, Van Houten et al., J. Biol. Chem. 261 [1986], 14135-14141), or by binding to the 5'- End, during solid phase oligonucleotide synthesis,  as a phosphoamidite derivative (U. Pieles and U. English, Nucleic acids res. 17 [1989], 285-299), or also after Completion of the synthesis to the fully protected Oligonucleotide (B.L. Lee et al., Biochem. 27 [1988], 3197- 3203).

The object of the present invention is to provide new nucleotide derivatives modified with psoralen, those during the solid phase synthesis of oligonucleotides the installation of the psoralen modification at any desired Allow position in oligonucleotide to pass through the thus available terms in 3'- and 5'-position a wider range of applications for this class of compounds to enable. This task is the subject of present invention solved.

Die im Verfahren zur Herstellung der Verbindungen der Formel I verwendeten neuen Verbindungen der Formel IIIThe in the process for the preparation of the compounds of formula I used new compounds of formula III

G-L-H²-Ps (III)G-L-H²-Ps (III)

Gegenstand der Erfindung sind deshalb auch die Verbindungen der Formel III, ihre Verwendung als Zwischenprodukt zur Synthese der Verbindungen der Formel I, und ein Verfahren zu ihrer Herstellung.wherein Ps, H², G and L have the meaning given above, represent a crucial intermediate for manufacturing of the compounds of formula I according to the invention with which the introduction of the psoralen remnant is effected. The invention therefore also relates to the compounds  of formula III, their use as an intermediate for Synthesis of compounds of formula I, and a method for their manufacture.

Of the compounds of formula III, those are compounds preferred in which H² = O and L tetramethylene, Hexamethylene and especially pentamethylene.

The process for the preparation of the compounds of formula III is characterized in that 4,5 ', 8-trimethylpsoralen reacted with bromomethyl methyl ether and the obtained 4'-bromomethyl-4,5 ', 8-trimethylpsoralen with one compound HO-L-H²-H, wherein L and H² are those given above Meanings, especially the preferred meanings, have, to a compound of formula HO-L-H²-Ps implements, and then this connection, for. B. by implementation with p-toluenesulfonyl chloride, in a manner known per se Transfer compound of formula III.

The reaction of 4,5 ′, 8-trimethylpsoralen with the bromomethyl methylether can be analogous to the instructions from Isaacs et al. (Biochem. 16 [1977], 1058-1064) take place; instead of highly toxic, carcinogenic and not commercially available According to the invention, chloromethyl methyl ether becomes commercially available available, considerably less toxic bromomethyl methyl ether used.

Die Einführung des Spacerarms (-H²-L-O-) kann wie bei U. Pieles und U. Englisch, Nucleic acids res. 17 (1989), 285-299, beschrieben, durchgeführt werden. The introduction of the spacer arm (-H²-L-O-) can be done as for U. Pieles and U. English, Nucleic acids res. 17 (1989), 285-299.

The reaction with para-toluenesulfonyl chloride (tosyl chloride) can be carried out in a manner known per se, preferably with pyridine as solvent.

Die erfindungsgemäßen Verbindungen der Formel I, insbesondere die bevorzugt genannten Verbindungen der Formel I, und in erster Linie die Verbindungen der Formel I, worin B die vorstehend genannte Gruppierung (1) darstellt, und die Reste bedeuten: X⁴ = H; X⁵ = Benzoyl; n = 1, H¹ = S; H² = O; L = Tetramethylen, Hexamethylen und insbesondere Pentamethylen; X¹ = H; X² =The compounds of formula I according to the invention, in particular the preferred compounds of the formula I, and primarily the compounds of formula I, wherein  B represents the above grouping (1), and the residues mean: X⁴ = H; X⁵ = benzoyl; n = 1, H¹ = S; H² = O; L = tetramethylene, hexamethylene and in particular Pentamethylene; X¹ = H; X² =

R⁴ = isopropyl; R⁵ = methyl or β-cyano-ethyl; X³ = 4,4′- Dimethoxytrityl; and Ps the meaning given above owns; are suitable as psoralen-modified nucleotide Building blocks very good for installing the psoralen modification in any position of oligonucleotides during their solid phase synthesis. Due to the non-binding terms in 5'- and 3'-position have the use of the invention Psoralen modified nucleotide building blocks synthesized oligonucleotides versus psoralen-modified Prior art oligonucleotides (see e.g. U. Pieles and U. English, Nucleic acids res. 17 [1989], 285-299, and B.L. Lee et al., Biochem. 27 [1988], 3197-3203) a much wider range of applications. The free terms allow further modifications, e.g. B. the connection of Biotin groups, metal chelates, etc., to the free termini. An enzymatic phosphorylation in the 5'-position enables the incorporation of the psoralen-modified oligonucleotides in larger DNA fragments and their radioactive labeling, and leads to further new applications, e.g. B. in affinity chromatography (the covalent linkage double-stranded DNA sections, coupled to a solid carrier, enables high salt concentrations in the elution of the desired protein without loss of capacity of the column material) and as an anti-sense oligonucleotide when examining structural details in DNA's and RNA's (which are due to the residues X² of the formula I modified internucleotide binding leads e.g. B. to one Increase in nuclease stability).

In their capacity as anti-sense oligonucleotides using the psoralen modified nucleotide building blocks synthesized oligonucleotides (e.g. those described in Example 9 below Psoralen-modified strands of the oligonucleotide pairs 1 to 5) due to the blocking effect on the Translation also a pronounced antiviral (virocidal) Effect (see e.g. Y. Shi et al., Nucleic acids res. 15 [1987], 6843-6854); they are therefore used as antivirals Chemotherapeutic, especially in AIDS therapy.

The invention therefore also relates to the use of compounds of the formula I according to the invention in the Solid phase synthesis of oligonucleotides for introduction of nucleotide building blocks modified with psoralen.

Another subject are those using the invention Compounds of formula I synthesized Psoralen-modified oligonucleotides and their use in affinity chromatography, as anti-sense Oligonucleotide and as an antiviral chemotherapy drug.

Beispiel 9Example 9 Photoreaktionen von unter Verwendung der Verbindung (VIII) nach Beispiel 8 modifizierter OligonucleotidePhotoreactions using compound (VIII) Oligonucleotides modified according to Example 8

Alle Oligonucleotide wurden mittels eines DNA-Synthesizers der Firma Applied Biosystems, Modell 380 B, unter Standardbedingungen durchgeführt (vgl. Nucleic Acids Res. 12 [1984], 4539-4557). All oligonucleotides were made using a DNA synthesizer from Applied Biosystems, model 380 B, under standard conditions carried out (cf. Nucleic Acids Res. 12 [1984], 4539-4557). For the modified nucleotides instead of tetrazole Nitrophenyltetrazole used, with one on 8 minutes extended coupling time. After the synthesis was completed the deprotection and purification of the Oligonucleotides again according to standard conditions.

The photo experiments were carried out in a fluorescence spectrometer made by Aminco, model 95. It was Irradiated for 50 minutes at a wavelength of 345 nm.

Only the unmodified one was used for the photo experiments Matrix strand at the 5'-end with T4 polynucleotide kinase and (γ³²P) -ATP marked under standard conditions. 300 pmoles of Oligonucleotides were in a total volume of 20 ul phosphorylated; in the same concentration modified strands dissolved in the kinization buffer, however without the addition of polynucleotide kinase and radioactivity.

Die Analyse der Reaktionsprodukte erfolgte mittels Autoradiographie.2 µl of the unmodified, radiolabelled matrix strand and 6 ul of the modified strand were in one Total volume of 20 µl mixed, at 85 ° C for 10 minutes heated and then left at 37 ° C for 20 minutes. This process serves to correctly hybridize the oligonucleotides. The hybridized oligonucleotides were then transferred to a quartz test tube and the sample Irradiated for 50 minutes at a wavelength of 345 nm. Then 8 µl formamide color mix (95% deionized formamide, 0.1% bromophenol blue, 0.1% xylene cyanol) added, the samples heated to 85 ° C. for 10 minutes, quickly cooled on ice and on a 20%, 8M Denaturing polyacrylamide gel containing urea applied. The analysis of the reaction products was carried out using autoradiography.

The following pairs of oligonucleotides were examined (with * the psoralen modification is marked):

Fig. 2 zeigt schematisch den Ablauf der Photoreaktion. Fig. 2 shows schematically the course of the photoreaction.

In FIGS. 3 and 4 are diagrams are shown of the gel analysis.

Fig. 3 is a schematic representation of the gel analysis (autoradiogram) shows the photoreaction of the pair of oligonucleotides 1 (Hybrid 1). Track 1 shows the control (without photoirradiation), track 1 a shows the same sample after 50 minutes of irradiation at 345 nm, track 1 c and 1 d each show the radiolabelled matrix or modified strand alone.

Fig. 4 shows the schematic representation shows the gel analysis of the photoreactions of pairs of oligonucleotides 2 to 5. The photoreactions are identified with "a" (50 minutes at 345 nm). The control samples are labeled without "a" (treated under the same conditions but without photoirradiation). From the extent of the resulting photoaddition products can be in lane 2 a and 3 a is a clear preference of the photoreactions of AT or recognize TA-positions, wherein each of the adenosine psoralen-modified is (yield the photoaddition more than 90%). In track 4 a, the reactive AT position was shifted by one base pair; the weak photoreaction clearly documents that a spacer length (L) of 5 carbon atoms represents the optimum. Track 5 a shows the possibility of a thymidine monoadduct or a crosslink between thymidine and cytidine which is also possible. The weak photoaddition shows that these reactions are possible.
